    Water quality monitoring and hydraulic evaluation of a household roof runoff harvesting system in France

    The quality of harvested rainwater used for toilet flushing in a private house in the south-west of France was assessed over a one-year period. Twenty-one physicochemical parameters were screened using standard analytical techniques. The microbiological quality of stored roof runoff was also investigated and total flora at 22°C and 36°C, total coliforms, Escherichia Coli, enteroccocci, Cryptospridium oocysts, Giardia cysts, Legionella species, Legionella pneumophila, Aeromonas, and Pseudomonas aeruginosa were analysed. Chemical and microbiological parameters fluctuated during the course of the study, with the highest levels of microbiological contamination observed in roof runoffs collected during the summer. Overall, the collected rainwater had a relatively good physicochemical quality but variable, and, did not meet the requirements for drinking water and a microbiological contamination of the water was observed. The water balance of a 4-people standard family rainwater harvesting system was also calculated in this case study. The following parameters were calculated: rainfall, toilets flushing demand, mains water, rainwater used and water saving efficiency. The experimental water saving efficiency was calculated as 87 %. The collection of rainwater from roofs, its storage and subsequent use for toilet flushing can save 42 m3 of potable water per year for the studied system

    Monitoring of water quality from roof runoff: Interpretation using multivariate analysis

    The quality of harvested rainwater used for toilet flushing in a private house in the south-west of France was assessed over a one-year period. Temperature, pH, conductivity, colour, turbidity, anions, cations, alkalinity, total hardness and total organic carbon were screened using standard analytical techniques. Total flora at 22°C and 36°C, total coliforms, Escherichia coli and enterococci were analysed. Overall, the collected rainwater had good physicochemical quality but did not meet the requirements for drinking water. The stored rainwater is characterised by low conductivity, hardness and alkalinity compared to mains water. Three widely used bacterial indicators - total coliforms, E. coli and enterococci - were detected in the majority of samples, indicating microbiological contamination of the water. To elucidate factors affecting the rainwater composition, principal component analysis and cluster analysis were applied to the complete data set of 50 observations. Chemical and microbiological parameters fluctuated during the course of the study, with the highest levels of microbiological contamination observed in roof runoffs collected during the summer. Escherichia coli and enterococci occurred simultaneously, and their presence was linked to precipitation. Runoff quality is also unpredictable because it is sensitive to the weather. Cluster analysis differentiated three clusters: ionic composition, parameters linked with the microbiological load and indicators of faecal contamination. In future surveys, parameters from these three groups will be simultaneously monitored to more accurately characterise roof collected rainwater

    Soil type and soil preparation influence vine development and grape composition through its impact on vine water and nitrogen status

    The influence of soil type and preparation on vine development and grape composition was investigated in a 50 ha estate located in Saint-Emilion (Bordeaux, France) and planted predominantly with Merlot. Part of the vineyard was planted down the slopes and another part of the vineyard was planted on terraces, where soils were profoundly modified through soil preparation. Grape composition (berry weight, sugar, total acidity, malic acid and pH), vigor (pruning weight), vine nitrogen status (Yeast Available Nitrogen (YAN) in grapes) and vine water status (δ13C) was measured at a very high density grid of 10 data points per hectare. Water deficit was globally weak over the estate because of high soil water holding capacity whereas vine nitrogen status was highly variable. Vine vigor and grape composition were predominantly driven by vine nitrogen status. On terraces, where soils were deep, due to invasive soil preparation, water deficits were particularly small or non-existent and vine nitrogen status was highly variable. Grape quality potential was medium to low, except in places with low nitrogen status, but at the expense of low yields. On parcels planted down the slopes water deficits were recorded because vine rooting was limited by compact subsoils. Vine nitrogen status was homogeneous. Grape quality and yield were medium to high and relatively homogeneous. When possible, downhill plantations are to be preferred over terraces because in the latter vine yield and quality parameters are highly variable because of massive soil movements prior to plantation

    Pesticides in roof runoff: Study of a rural site and a suburban site

    The quality of stored roof runoff in terms of pesticide pollution was assessed over a one-year period. Two tanks, located at a rural and suburban site, respectively, were sampled monthly. The two studied collection surface were respectively a tile slope roof and a bituminous flat roof. Four hundred and five compounds and metabolites were screened using liquid and gas chromatography coupled with various detection systems. Principal Component Analysis was applied to the data sets to elucidate patterns. At the rural site, two groups of compounds associated with two different types of agriculture, vineyard and crops, were distinguished. The most frequently detected compound was glyphosate (83%) which is the most commonly used herbicide in French vineyards. At the suburban site, quantified compounds were linked to agriculture rather than urban practices. In addition, all samples were contaminated with mecoprop which is a roof-protecting agent. Its presence was attributed to the nature of roofing material used for rainwater collection. For both sites, the highest number and concentrations of compounds and metabolites were recorded at the end of spring and through summer. These results are consistent with treatment periods and higher temperatures

    Insights into the Biodiversity, Behavior, and Bioluminescence of Deep-Sea Organisms Using Molecular and Maritime Technology

    Since its founding, the Monterey Bay Aquarium Research Institute (MBARI) has pioneered unique capabilities for accessing the deep ocean and its inhabitants through focused peer relationships between scientists and engineers. This focus has enabled breakthroughs in our understanding of life in the sea, leading to fundamental advances in describing the biology and the ecology of open-ocean and deep-sea animals. David Packard’s founding principle was the application of technological advances to studying the deep ocean, in part because he recognized the critical importance of this habitat in a global context. Among other fields, MBARI’s science has benefited from applying novel methodologies in molecular biology and genetics, imaging systems, and in situ observations. These technologies have allowed MBARI’s bioluminescence and biodiversity laboratory and worldwide collaborators to address centuries-old questions related to the biodiversity, behavior, and bio-optical properties of organisms living in the water column, from the surface into the deep sea. Many of the most interesting of these phenomena are in the midwater domain—the vast region of ocean between the sunlit surface waters and the deep seafloor

    Crosstalks between Myo-Inositol Metabolism, Programmed Cell Death and Basal Immunity in Arabidopsis

    BACKGROUND: Although it is a crucial cellular process required for both normal development and to face stress conditions, the control of programmed cell death in plants is not fully understood. We previously reported the isolation of ATXR5 and ATXR6, two PCNA-binding proteins that could be involved in the regulation of cell cycle or cell death. A yeast two-hybrid screen using ATXR5 as bait captured AtIPS1, an enzyme which catalyses the committed step of myo-inositol (MI) biosynthesis. atips1 mutants form spontaneous lesions on leaves, raising the possibility that MI metabolism may play a role in the control of PCD in plants. In this work, we have characterised atips1 mutants to gain insight regarding the role of MI in PCD regulation. METHODOLOGY/PRINCIPAL FINDINGS: - lesion formation in atips1 mutants depends of light intensity, is due to PCD as evidenced by TUNEL labelling of nuclei, and is regulated by phytohormones such as salicylic acid - MI and galactinol are the only metabolites whose accumulation is significantly reduced in the mutant, and supplementation of the mutant with these compounds is sufficient to prevent PCD - the transcriptome profile of the mutant is extremely similar to that of lesion mimic mutants such as cpr5, or wild-type plants infected with pathogens. CONCLUSION/SIGNIFICANCE: Taken together, our results provide strong evidence for the role of MI or MI derivatives in the regulation of PCD. Interestingly, there are three isoforms of IPS in Arabidopsis, but AtIPS1 is the only one harbouring a nuclear localisation sequence, suggesting that nuclear pools of MI may play a specific role in PCD regulation and opening new research prospects regarding the role of MI in the prevention of tumorigenesis. Nevertheless, the significance of the interaction between AtIPS1 and ATXR5 remains to be established

    Effect of angiotensin-converting enzyme inhibitor and angiotensin receptor blocker initiation on organ support-free days in patients hospitalized with COVID-19

    IMPORTANCE Overactivation of the renin-angiotensin system (RAS) may contribute to poor clinical outcomes in patients with COVID-19. Objective To determine whether angiotensin-converting enzyme (ACE) inhibitor or angiotensin receptor blocker (ARB) initiation improves outcomes in patients hospitalized for COVID-19. DESIGN, SETTING, AND PARTICIPANTS In an ongoing, adaptive platform randomized clinical trial, 721 critically ill and 58 non–critically ill hospitalized adults were randomized to receive an RAS inhibitor or control between March 16, 2021, and February 25, 2022, at 69 sites in 7 countries (final follow-up on June 1, 2022). INTERVENTIONS Patients were randomized to receive open-label initiation of an ACE inhibitor (n = 257), ARB (n = 248), ARB in combination with DMX-200 (a chemokine receptor-2 inhibitor; n = 10), or no RAS inhibitor (control; n = 264) for up to 10 days. MAIN OUTCOMES AND MEASURES The primary outcome was organ support–free days, a composite of hospital survival and days alive without cardiovascular or respiratory organ support through 21 days. The primary analysis was a bayesian cumulative logistic model. Odds ratios (ORs) greater than 1 represent improved outcomes. RESULTS On February 25, 2022, enrollment was discontinued due to safety concerns. Among 679 critically ill patients with available primary outcome data, the median age was 56 years and 239 participants (35.2%) were women. Median (IQR) organ support–free days among critically ill patients was 10 (–1 to 16) in the ACE inhibitor group (n = 231), 8 (–1 to 17) in the ARB group (n = 217), and 12 (0 to 17) in the control group (n = 231) (median adjusted odds ratios of 0.77 [95% bayesian credible interval, 0.58-1.06] for improvement for ACE inhibitor and 0.76 [95% credible interval, 0.56-1.05] for ARB compared with control). The posterior probabilities that ACE inhibitors and ARBs worsened organ support–free days compared with control were 94.9% and 95.4%, respectively. Hospital survival occurred in 166 of 231 critically ill participants (71.9%) in the ACE inhibitor group, 152 of 217 (70.0%) in the ARB group, and 182 of 231 (78.8%) in the control group (posterior probabilities that ACE inhibitor and ARB worsened hospital survival compared with control were 95.3% and 98.1%, respectively). CONCLUSIONS AND RELEVANCE In this trial, among critically ill adults with COVID-19, initiation of an ACE inhibitor or ARB did not improve, and likely worsened, clinical outcomes.     Usages problématiques d'Internet : un état des lieux de la fracture numérique de troisième niveau en France

    La politique publique d'accompagnement aux usages numériques s'est implicitement fondée sur la réduction des fractures numériques de premier et de deuxième niveau. D'abord en facilitant l'accès (équipement et infrastructure) puis en se préoccupant des usages ou des non-usages. Même si l'illectronisme reste important en France, l'éducation aux usages est aujourd'hui un axe privilégié par les autorités publiques. Nous testons l'existence d'une nouvelle fracture numérique, que nous qualifions de troisième niveau, et qui concerne les usages problématiques de l'Internet observés sur un échantillon représentatif de la population française en 2020. 10 à 14% de notre échantillon est concerné et cela révèle une intrication forte entre un usager et des usages mais aussi des effets ambivalents sur la santé mentale, la vie sociale et professionnelle. Cette troisième fracture appellerait à repenser une politique publique d'accompagnement garantissant un usage sobre et modéré et le développement de modèle économique du numérique plus vertueux et orienté « well-being by design »
